The top-seeded American twins went down to US-South African pair Eric Butorac and Raven Klaasen 7-6 (11/9), 6-4 in the third round.
The defeat ends their bid for a seventh title at the Australian Open, their favourite Grand Slam tournament.
The 35-year-old twins have a record 15 Grand Slam doubles titles, ahead of Australian pair John Newcombe and Tony Roche on 12.
